Copper induces expression and methylation changes of early development genes in Crassostrea gigas embryos ArchiMer

Copper contamination is widespread along coastal areas and exerts adverse effects on marine organisms such as mollusks. In the Pacific oyster, copper induces severe developmental abnormalities during early life stages; however, the underlying molecular mechanisms are largely unknown. This study aims to better understand whether the embryotoxic effects of copper in Crassostrea gigas could be mediated by alterations in gene expression, and the putative role of DNA methylation, which is known to contribute to gene regulation in early embryo development. Crassostrea gigas ferritin: cDNA sequence analysis for two heavy chain type subunits and protein purification ArchiMer

Ferritin has been shown as being the principal iron storage in the majority of living organisms. In marine species, ferritin is also involved in high-level accumulation of Po-210. As part of our work on the investigation of these radionuclides' concentration in natural environment, ferritin was searched at the gene and protein level. Ferritin was purified from the visceral mass of the oyster Crassostrea gigas by ion-exchange chromatography and HPLC. SDS-PAGE revealed one band of 20 kDa. This study presents the main results of oyster growth obtained experimentally in 2002 and 2003, in the farming area of Cancale before the leases reorganization of 2004. According to the REMORA monitoring network from 1990 to 2004, the oyster growth in Cancale bay may be characterized by an average annual weight gain (around 30g per year), a rather weak meat content compared to other regional oyster sites, and an exceptional quality of shell (absence of blisters, few Poly do rd). A study on mortalities, growth and fattening of the oyster Crassostrea gigas was carried out spring with the autumn 1987 in four sites of bay of Bourgneuf. Mortalities, more important in spring, prove to be higher in the sectors North compared to the Southern sectors of bay of Bourgneuf. The performances of growth are rather weak when they are compared with other French oyster sectors. They however present a better production in the cattle-breeding areas located at the South of bay. Cryopreservation is widely used for long-term conservation of various tissues, embryos or gametes. However, few studies have described cryopreservation of invertebrate primary cell cultures and more particularly of marine invertebrate somatic cells. This technique would however be of great interest to facilitate the study of various metabolic processes which vary seasonally. Pacific oyster Crassostrea gigas is one major species for aquaculture, and the development of breeding programs and the need for preservation of wild stock genetic resources prompted the need for larvae cryopreservation. The objective of the present study was to choose the most reliable protocol from several existing publications, to test its biological and practical limitations, and to adapt it to hatchery conditions. In the present work, cultured atrial cells isolated from oyster (Crassostrea gigas) were used as an experimental model to investigate the toxicity of tributyltin (TBT) after short-time exposure at concentrations representative of those that can be measured in seawater, marine sediments and/or bivalves bioaccumulating this pollutant. Cytotoxic and antibacterial properties of Mytilus galloprovincialis, Ostrea edulis and Crassostrea gigas (bivalve molluscs) hemolymph ArchiMer

Mussel (Mytilus galloprovincialis) plasma contains cytotoxic activity against both vertebrate (erythrocytes and mouse tumour) and protozoan cells. Procaryotes (Escherichia coli and Vibrio alginolyticus) were not sensitive to the cytotoxicity. The activity was still present in dialyzed samples but was inhibited by heating at 45°C. Delivery of riboflavin to larval and adult pacific oysters, Crassostrea gigas Thunberg by lipid spray beads ArchiMer

Lipid spray beads (SB) were prepared containing 13% w/w particulate riboflavin. Beads suspended in seawater lost 73% riboflavin after 24 h. Release of riboflavin from SE ingested by Pacific oyster (Crassostrea gigas) larvae was observed under epifluorescent light. Riboflavin concentrations in tissues of adult oysters fed on riboflavin-SB were significantly (SNK, P < 0.05) greater than those of oysters fed on seawater-filled SE. Concentrations of riboflavin in oysters exposed to dissolved riboflavin were not significantly greater than those of oysters fed on seawater-filled SE: indicating that elevated riboflavin concentrations in oysters fed on riboflavin-SB were attributable to breakdown of ingested beads rather than uptake of dissolved riboflavin...

The prophenoloxidase (ProPO) system is the origin of melanin production and is considered to be an innate defence mechanism in invertebrates. In different bivalve species, phenoloxidase (PO) is present in the haemolymph as an inactive form of ProPO. The present study focuses on the Pacific adult oyster, Crassostrea gigas, an economically important bivalve species along French coasts. The results indicate that many factors may inhibit the PO-like activity. Ostreid herpesvirus 1 (OsHV-1) infections have been reported around the world and are associated with high mortalities of the Pacific oyster (Crassostrea gigas). In the summer 2008, abnormal mortality rates ranging from 80% to 100% were reported in France and affected only Pacific oysters. Analyses of oyster samples collected during mortality outbreaks demonstrated a significant detection of OsHV-1 (75% of analysed batches), which appeared stronger than previous years. Detection of ostreid herpesvirus-1 (OsHV-1) by PCR using a rapid and simple method of DNA extraction from oyster larvae ArchiMer

A DNA extraction procedure was developed for the detection of ostreid herpesvirus-1 (OsHV-1) using the polymerase chain reaction (PCR) in oyster larvae. The DNA extraction procedure developed was tested on 8 larval samples. Abnormal nuclei with characteristic features associated with OsHV-1 infections were only observed in samples in which the viral DNA was detected by PCR. A previously described competitive PCR method was applied to detect inhibition during PCR reactions. The results show that the method can be used on small amounts of oyster larvae (3 mg) for the detection of OsHV-1 DNA by PCR.

The development of molecular diagnostic assays with increased sensitivity compared with conventional histological techniques is highly desirable for effective management of bonamiosis in cultured oyster stocks and wild populations. A real-time TaqMan PCR assay was developed for the specific detection of Bonamia species in infected oyster tissues. The TaqMan assay was shown to be significantly more sensitive than histopathology. Although a real-time TaqMan PCR assay is comparable with conventional PCR in terms of sensitivity, it offers the advantages that it is a rapid test and has a very low risk of sample cross-contamination.
